- Discuss the sort in advance with the facility staff. Here you can find the minimum information needed:
- the type of cells to be collected
- the stain you will be using
- temperature of acquisition
- whether you want to collect into tubes, plates or slides
- number of populations you want to sort
- Samples must be in a single cell suspension. Clumps and debris can clog the fluidic of the instrument. It is essential to use filters for isolate cells from aggregates and debris.
- Always keep your cells on ice to stop cell death.
- If you are using a single stain sample, bring with you a tube of unstained cells (ideally 60,000-300,000 cells) in order to clearly define the negative cell population. If you are using more fluorochromes, bring with you the unstained sample and FMO controls (fluorescence minus one).
